Quickest and best value way will undoubtfully be train. There are daily several trains passing Nanjing and going to Hangzhou. Travel time for fastest trains : 5 to 6 hours. Check http://www.travelchinaguide.com/china-trains/index.htm

You can take the long-distance coach at the following three long-distance bus stations: Nanjing Eastern Long-Distance Bus Station (Nanjing Changtu Qiche Dongzhan) Nanjing Hanfu Street Passenger Transport Bus Station (Nanjing Hanfujie Keyunzhan) Nanjing Hanzhong Gate Long-Distance Bus Station (Nanjing Hanzhongmen Changtu Qichezhan) The ticket can be bought either at the station or at Nanjing Long-Distance Bus Station(Zhong Yang Gate).

For a flight leaving at 1410 you'd need to be at Hangzhou airport at 1210. I would allow about 1 hour to get from downtown Hangzhou to the airport, so you'd need to arrive in Hangzhou by 1100. Seeing that it usually takes about 3.5 hours for the Nanjing -- Hangzhou bus, I would catch a bus leaving Nanjing before 0730. The possible departures are therefore 0620, 0700 and 0720.

There is no train from Nanjing to Yixing or from Yixing to Hangzhou. It seems there is a bus station called Dingshan(丁山) in Dingshuzhen, which has buses running to Hangzhou. But you'd better make a confirmation after arriving at Dingshuzhen. Otherwise, you need to retact to Yixing to take the bus to Hangzhou.
