I am a U.S citizen studying abroad in Singapore and I want to visit my friend in Shanghai three times. I am planning on applying for my Chinese visa in Singapore. Will it be possible to get a multiple entry L visa?

You mentioned you study in Singapore, so I guess you have been staying there for a long time. If so, it's possible to apply for multiple entry L visa but it cannot be assured.

I am traveling to China for the second time in a year, but to different places. I have a valid "L" visa with "M" entries. Do I need to provide an "Invitation letter" again or am I OK without doing that?

You don't need to provide an Invitation Letter. You can simply enter China with your L visa.
