There is a ferry from Macau Maritime Ferry Terminal to Guangzhou Nansha Port on Saturday and Sunday. But it is not operated regularly. I suggest you contact them to gain the details: 020-84688963、84688716

Ferry ticket: CNY 180 Duration: around 80 minutes.

If not, you can get through Gongbei Border to reach Zhuhai and take long distance bus at Gongbei Bus Station.

Your wife do not need a visa in Macau but she has to apply for a visa if she wants to enter Guangzhou. She can apply her visa at Chinese embassy in Thailand or just find the CTS(China Travel Service) in Macau to help her get a visa. CTS is very capable but you have to pay for service fee.

From website we are informed that Singaporean will be given visa free for 15 days on social visit. You may enquire at the China Consulate for confirmation.
